Western Blotting Analysis (5 seconds exposure)
- Wash membrane-bound HRP (antibody conjugate) three times for 5 minutes in TBS/T.
- Prepare 1X ECL Western Blotting Substrate by mix Reagent A and Reagent B at a 1000:5 ratio (i.e., 5mL Reagent A + 25µL Reagent B for 6 x 8 cm² membrane) and add it to the blot.
- Incubate Reagent with membrane for 1 minute, remove excess solution (membrane remains wet), wrap in plastic.
- Expose blot to X-ray film: XAR ALF 2025 or XAR ALF 1318
Western blot analysis of diluted HeLa cell lysates using anti- β-Actin antibody. Lane 1 contained 2µg of total protein, and four 2-fold serial dilutions were applied to lanes 2-5 at 10µL/well.
